5.6.1 General guidelines

• Make sure that the HPLC column is stable (in power and temperature) and at PH in your mobile phase.

• If the mobile phase is stored for a certain period, a high background current can be observed.

• When using the detector to analyze complex biological samples, the right sample preparation is especially important.

• Rinse new analysis columns to waste a few hours before they connect with the system.Use a special column for each specific analysis to prevent cross -contamination of the column.

• To fully use the possibilities of power connections in a UHPLC system, especially when using columns with smaller inner diameters, minimize dead volumes before the detector. The shortest possible length.

Tip: When performing a gradient, it is recommended to perform empty injections with the desired LC-methodgradient profile before analysis.

5.6.2 Setting a filter constant

The filter time constant is used to electronically reduce the noise of the basic line within chromatographic driving.

See section 5.3.3 (→ Page 50) For more information about setting the filter constant of the display.

5.6.3 Selection of the evaporation temperature

General remarks

The temperature -controlled evaporation pipe dries the aerosol from the nebulizer. Thus, the evaporation pipe removes the mobile phase components and leaves an aerosol with dried analytes. You have set the evaporation temperature, wait about 30 minutes to ensure that the evaporation pipe comes to balance before the sample analysis is started.

Tips: A change in the evaporation temperature can have a dramatic effect on the detector's response.

Corona VEO RS Only If you decide to use an optimized evaporation temperature, you must perform three or more injections of a reference standard for the analytical method.Suitable starting point temperature, based on the performance criteria as stated earlier.

5.6.4 Use gas flow states (Corona VEO RS only)

The Corona VEO RS detector is capable of two separate gas pressure states: analytical gas stream mode and microlc gas flow mode.

Each of these modes sets a specific pressure based on the standard gum pressure for the nebulizer. They are designed as a simple way to optimize the efficiency of the atomizer when using ML/min - 2.00 ml/min.

• Analytic gas flow status: The sentence point of the gas pressure corresponds to the standard gas pressure setting as described in the certificate sent with the nebulizer.

• Microlc gas flow mode: The gas pressure value is calculated by multiplying the analytical gas stream mode by a factor to optimize the gas pressure for certain types of liquid current speeds and mobile phase assemblies.

5.6.5 Application of a power function value

The signal from the detector has a wide dynamic range with more than four size orders.Linear interval.has been scaled and sent.

The choice of an optimal power function value will result in an increase in the linear dynamic range for the detector compared to the standard setting.

Tips: Changes in PFV do not lead to improvements in the limit of the detector for detection (LOD).

The standard PFV setting of 1.0 offers a good universal setting for most applications and connections.

The detector accepts each value between 0.67 and 2.00 in steps of 0.01.

To set the power function value of the detector display:

1. On the detector screen, select the main menu in the Run mode Soft key menu.

2. Set control parameters as required in your application.

3. For the Power function parameter, enter the three digits in the desired PFV without a decimal point.

4. Press Enter.databrømen is now being raised to the selected current.

5.6.6 Minimalization of baseline -inise

Minimalization of baseline judge generated for a certain LC method depends on many factors.

• Only use mobile phases of the highest available quality and keep impurities and pollutants to a minimum.

• Rinse the LC system periodically to remove non-volatile mobile phase residues if applicable.

• View the recommended maintenance plan for the detector and the LC system in accordance with the manufacturer's guidelines.

• The gas supply must be filtered and free of moisture and hydrocarbons.

Important: when connecting TTL inputs, Shell 0.5 A.

Important: when connecting TTL -inputs

TTL -input the inputs are TTL compatible and get "Gas off", "Autozero", "Start" and "Divert".One is used.Change of TTL level, the tension must be between 0 and 5 V and the unity of the feeling of polarity must be matched (ie "+" and "-" to the external unity must be connected "and"-"of the detector). The "+" port is the top of each group, " -" Poort is immediately below, followed by the earth (GND).

• Start the "Start" input is available for use with a chromatographic data system that requires the activation of eventsnchronization to data collection from an external device, such as an autosampler. At least 0.5 seconds) of the external device.

• Gas from the "gas from" input can be used to switch off the internal gas inlet valve and to stop the gas supply to the nebulizer.

If the "Gas OFF" input is enabled, the detector closes the internal gas supply.

• Autozero "Autozero" input can be used to set the signal of a channel on a "zero point", regardless of the measured power.

Allow enough time after you have carried out Autozero and before injection to ensure that the Autozero assignment is completed. It is expected that reasonably flat (ie 15 seconds before the elution of a top).

• Give the "diversion" input can be used to control the power -changing valve with a driver for the third party.

Relay outputs

"Pump OFF" output switch is used to send a signal to the pump (or other external device) to stop the power of mobile phase and switch off the pump. With a mobile phase if the gas pressure falls less than 50 % of the required gas volume for more than 60 seconds. To switch off the liquid flow.In some cases a special cable is required.

The pins with the "CC1" "" and "CC2" label offer contact closures that can be used to start an external device (for example, an autosampler) or to change a valve position. Output switches are used, connects the outputAccording to their polarity ("+" or "-").
